VitroGel® MSC is a xeno-free (animal origin-free) hydrogel system developed to support three-dimensional (3D) cultures of mesenchymal stem cells (MSCs).  This hydrogel system can be used to make hydrogel cell beads for MSC scale-up.  Microcarriers are not required for MSC scale-up.

VitroGel® MSC is ready-to-use with an optimized formulation that fully supports the rapid expansion of MSCs. Brilliant Sodium Assays
 
ION’s Brilliant Sodium Assay is a total assay solution for multi-well plate-based, high-throughput measurements of changes in intracellular Na+ mediated through a wide-variety of plasma membrane and intracellular sodium channels and transporters. In multi-well, plate-based formats, the Brilliant Sodium Assay can be used to discover and characterize the effects of many tens-of-thousands of compounds and environmental factors on effectors of intracellular Na+. ION’s Brilliant Sodium Assay provides all the reagents necessary for use as a wash or no-wash assay with adherent or non-adherent cells. The optional use of a probenecid solution and an extracellular background masking solution (TRS) offers the ultimate in compatibility for cells types which are difficult to load with fluorescent Na+ indicators (e.g. Chinese Hamster Ovary, CHO cells) and when performing assays in complete, serum-containing cell culture medium is desired.

The enzyme-sensitive hydrogel can be degraded by several MMPs (MMP1, MMP2, MMP3, MMP9, and MMP13) and support different biological activities such as cell proliferation, migration (adhesion/dispersion), differentiation, angiogenesis, apoptosis, etc.  VitroGel® MMP High Concentration comes with VitroGel® Dilution Solution to adjust the final hydrogel strength from 10 to 4000 Pa.

ION Biosciences PBFI AM
 
PBFI is a UV-excitable, green ratiometric indicator for intracellular potassium (K) measurements. Ex/Em: 340/505 nm can be used to measure K-bound PBFI, and Ex/Em: 380/505 nm can be used to detect K-free PBFI. It is 1.5X more selective for K over Na, and works well in cytosolic conditions where [K]/[Na] is ~10. Ratiometry is optimal for imaging applications where quantification of intracellular K concentrations is desired, and reduces effects of photobleaching, heterogenous dye loading, and variable cell morphology.

This hydrogel system is ready to use with an optimized formulation that fully supports the rapid expansion of high-quality 3D stem cell spheroids with pluripotent properties. hPSCs directly thawed from liquid nitrogen or passaged from 2D matrix coated culture vessels can be immediately mixed with the hydrogel solution for static suspension cultures. Moreover, the optimization protocol is ideal for time-sensitive experiments, as it does not require excessive medium exchanges, which can ultimately save on time and materials. ION Natrium Green – 2 (ING-2) is a yellow-green fluorescent, intracellular sodium (Na+) indicator with improved cellular loading and significantly higher brightness than SBFI. Its spectral properties (Ex/Em: 525 nm/545 nm) and large dynamic range make ING-2 the best Na+ indicator for high-throughput screening applications targeting Na+ channels, and non-selective monovalent cation channels. ING-2 has a more physiologically relevant affinity (Kd = 20 mM) than ING-1 (Kd = 92 mM). Also compatible with fluorescence microscopy using common fluorescein, GFP or more ideally YFP filters.

Reporter gene based Luminescent assay systems from Perkin Elmer provide a homogeneous assay with 96, 384, and 1536 well formats, no DTT (no smell formulation), Ease of storage (2 to 8°C) and shipping (at RT).
