Fluorescents, Dyes, and Assays
Essential Tools for Visualization, Detection, and Analysis
- Fluorescents include fluorophore-based reagents used for visualization, labeling, and fluorescence detection in workflows such as flow cytometry
- Dyes are used for conjugation, visualization, and molecular labeling applications across immunofluorescence, IHC, and in vivo workflows
- Assays are made to support quantitative and qualitative analysis in life science and molecular biology applications

PBFI is a UV-excitable, green ratiometric indicator for intracellular potassium (K) measurements. Ex/Em: 340/505 nm can be used to measure K-bound PBFI, and Ex/Em: 380/505 nm can be used to detect K-free PBFI. It is 1.5X more selective for K over Na, and works well in cytosolic conditions where [K]/[Na] is ~10. Ratiometry is optimal for imaging applications where quantification of intracellular K concentrations is desired, and reduces effects of photobleaching, heterogenous dye loading, and variable cell morphology.
Reporter gene based Luminescent assay systems from Perkin Elmer provide a homogeneous assay with 96, 384, and 1536 well formats, no DTT (no smell formulation), Ease of storage (2 to 8°C) and shipping (at RT). Different assay formats allow for varying levels of intensity and half-life depending on your application.
- Choose britelite plus:
- Half-life = 30 min
- Brightest signal = when looking for maximum sensitivity
- Shortest half-life = for continuous processing of samples
- Choose neolite:
- Half-life = 150 min
- High signal intensity, longer lasting signal = good balance
- For both continuous and batch processing of samples
- Best robustness (less dependent on mixing quality)
- Less sensitive to phenol red
- Can be stored 1 month at 22°C
- Choose steadylite plus:
- Half-life = 240 min
- Longest signal half-life = for batch processing of samples
- Super-sensitivity not needed
Perkin Elmer's ATPlite simple and sensitive system is an excellent kit to determine cell number due to proliferation or toxicity. It effectively replaces the MTT assay and can be used instead of the 3H thymidine assays with additional analysis.
- Choose ATPlite (2 steps):
- Half-life> 4 hours
- Longest half - life = for batch processing of samples
- 2 step system with separate lysis and detection steps
- More suited for Academia Choose
- ATPlite 1step:
- Measure within 30 min
- Brightest signal (~3x the counts of ATPlite 2Steps) = when looking for maximum sensitivity
- For continuous processing of samples
- True 1 step system = More suited to HTS
