Cell Culture Guidelines

  • Recommended seeding density: 1 x 103 to 1 x 105 cells/cm2
  • Initial adhesion phase (first 24 to 48 hours):
    • Rotation speed: 0.5 to 1.0 rpm
    • For cells with lower adhesion:
      • Rotation speed: 0.3 to 0.5 rpm

Cell Harvesting Protocol

  • Rinse the bottle twice with PBS
  • Add a dissociation solution (e.g., trypsin), ensuring full coverage of the bottle interior and pleats
  • Place the bottle in an incubator and rotate at 0.3 to 0.5 rpm for 5 to 10 minutes
  • After cell detachment, add an equal volume of cell culture medium to neutralize trypsin
  • Rotate the bottle to collect the cell suspension
  • Remove cells by pouring or pipetting
